天然蛋白A(Protein A)是一种发现于金黄色葡萄球菌的细胞壁表面蛋白,天然蛋白G(Protein G)是一种分离自G型或C型链球菌属的细胞表面蛋白,二者功能相似,主要通过与免疫球蛋白(Ig)的Fc区相互作用,可结合大多数哺乳动物的IgG。然而两者结合特异性上有所不同(详见说明书附录1,蛋白A,G对不同物种Ig的结合能力总表)。蛋白A,蛋白G常共价偶联在固体载体如琼脂糖珠或丙烯酸树脂小珠上,直接用以做免疫沉淀(IP)或者免疫共沉淀(Co-IP)来进行蛋白功能相关研究,或者直接装在层析柱上来纯化单克隆或者多克隆抗体。
本品使用的是基因改造后的蛋白A和蛋白G,不仅维持其本身的Ig亲和特性,同时也去除了天然蛋白本身的非主要结合域以降低非特异性结合。本品同时将蛋白A和蛋白G共价偶联到琼脂糖凝胶微球表面,比单独的蛋白A或者蛋白G都有更广的结合范围,适合于所有蛋白A琼脂糖珠和蛋白G琼脂糖珠单独可以结合的Ig,实用性更高。
